T4 DNA Polymerase - NEB
T4 DNA Polymerase catalyzes the synthesis of DNA in the 5´→ 3´ direction and requires the presence of template and primer. This enzyme has a 3´→ 5´ exonuclease activity which is much more active than that found in DNA Polymerase I ( E. coli ).

Thermo Scientific T4 DNA Polymerase is a template-dependent DNA polymerase that catalyzes 5'-3' synthesis from primed single-stranded DNA. The enzyme has a 3'-5' exonuclease activity, but lacks 5'-3' exonuclease activity.
Its two activities make T4 DNA Polymerase a useful enzyme for generating blunt ends on any duplex DNA molecule and for labeling DNA by a method known as replacement synthesis. This bulletin describes conditions for using T4 DNA Polymerase for both purposes.

T4 DNA polymerase is a mesophilic DNA polymerase that catalyzes 5′ → 3′ synthesis of DNA and has 3′ → 5′ exonuclease (proofreading) activity, but no 5′ → 3′ exonuclease activity. It removes 3′ overhangs and fills-in 5′ overhangs to form blunt ends.
T4 DNA Polymerase is purified from a T4 N82-infected E. coli by a modification of Challberg and Englund. This enzyme exhibits 5′→3′ DNA polymerase and 3′→5′ exonuclease activity (1,2,3). It may be used to fill restriction endonuclease termini and in replacement synthesis for the generation of radioactive probes (1,2).

T4 DNA Polymerase catalyzes the 5´→3´ synthesis of DNA from a primed single-stranded DNA template. Although possessing a potent 3´→5´ proofreading exonuclease, T4 DNA Polymerase contains no 5´→3´ exonuclease activity.
